Diabetes Impaired Ischemia-Induced PDGF (Platelet-Derived Growth Factor) Signaling Actions and Vessel Formation Through the Activation of Scr Homology 2-Containing Phosphatase-1

Objective: Critical limb ischemia is a major complication of diabetes characterized by insufficient collateral vessel development and proper growth factor signaling unresponsiveness. Although mainly deactivated hypoxia, phosphatases are important players in the deregulation proangiogenetic pathways. Previously, SHP-1 (Scr homology 2-containing phosphatase-1) was found to be associated with downregulation actions diabetic muscle. Thus, we aimed gain further understanding impact on smooth muscle cell (SMC) function under hypoxic conditions. Approach Results: Despite being inactivated conditions, high glucose level exposure sustained phosphatase activity SMC increased its interaction PDGFR (platelet-derived receptor)-β, thus reducing PDGF proangiogenic actions. Overexpression an inactive form fully restored PDGF-induced proliferation, migration, pathways exposed hypoxia. Nondiabetic mice deletion specifically were generated. Ligation femoral artery performed, blood flow measured for 4 weeks. Blood reperfusion, vascular density maturation, survival all improved while apoptosis attenuated SMC-specific null as compared mice. Conclusions: Diabetes maintained preventing hypoxia-induced SMC. Specific partially reperfusion ischemic limb. Therefore, local modulation could represent potential therapeutic avenue improve properties diabetes.
